چکیده
Amylases are key enzymes in the processing of starch many kingdoms life. They important catalysts industrial biotechnology where they applied in, among others, food and production detergents. In man amylases first digestion to glucose arguably also preferred target therapeutic strategies aimed at treatment type 2 diabetes patients through down-tuning assimilation. Efficient sensitive assays that report selectively on retaining amylase activities irrespective nature complexity biomaterial studied great value both finding new effective human inhibitors discovery microbial with potentially advantageous features for biotechnological application. Activity-based protein profiling (ABPP) glycosidases is inherently suited development such an assay format. We here design synthesis 1,6-epi-cyclophellitol-based pseudodisaccharides equipped a suite reporter entities their use ABPP from saliva, murine tissue as well secretomes fungi grown starch. The activity efficiency probes substantiated by extensive biochemical analysis, selectivity over related endoglycosidases validated structural studies.

Top-Down Approach to Protein Profiling 10s–100s of Complex Biological Samples
in 2006 to provide researchers engaged in biomarker discovery with a high-throughput, high-sensitivity approach to protein expression profiling. The ProteinChip system incorporates chip-based applications with top-down mass spectrometry (MS) for protein profiling, an area of research that has typically relied on more conventional approaches such as 2-D gel electrophoresis. Since its introductio...

Protein microarray technology has been successfully used for identifying substrates of purified activated kinases. We used protein microarrays to globally interrogate the effects of PTEN and Akt activity on the phospho-kinome of in vitro and in vivo glioma models and validated results in clinical pathological specimens. Whole cell lysates extracted from tumor samples can be applied to human kin...
متن کاملCharacterization of α-Synuclein Multimer Stoichiometry in Complex Biological Samples by Electrophoresis
The aberrant aggregation of α-synuclein in the brain is a hallmark of Parkinson's disease (PD). In vivo soluble α-synuclein occurs as a monomer and several multimers, the latter of which may be important for the biological function of α-synuclein. Currently, there is a lack of reproducible methods to compare α-synuclein multimer abundance between complex biological samples. Here we developed a ...
متن کاملGlycosylation and secretion of human α-amylases
Three human α-amylases exist: Amy1 (salivary amylase), Amy2A (pancreatic amylase), and Amy2B (expressed in various tissues). These amylases share a 97% 99% amino acid sequence identity, and two potential N-glycosylation sites (N427 and N476) are commonly found in the C-terminal region. In general, salivary amylase is more frequently glycosylated than pancreatic amylase, and it is still uncertai...
متن کاملActivity-based protein profiling: the serine hydrolases.
With the postgenome era rapidly approaching, new strategies for the functional analysis of proteins are needed. To date, proteomics efforts have primarily been confined to recording variations in protein level rather than activity. The ability to profile classes of proteins on the basis of changes in their activity would greatly accelerate both the assignment of protein function and the identif...
